In LDD, you were able to draw a line and then project it to a surface. The line would then take the elevation info from the surface and adjust the line as necessary, the same as sampling ti for a profile.
Can this also be done in Civil 3D 2010?

I think (not completely sure) you are looking for the Create Feature Lines from Objects found under the Grading menu or can be called from the command line with the _AeccCreateFeatureLines command.
Or maybe select the line, right-click -> Quick Profile...?

You'll want to use the Elevation From Surface.

Yes.
1. Draw a polyline.
2. Use the _AeccFeatureElevsFromSurf command (it's also found on the Modify tab, Edit Elevations panel, in a small toolbar button)
