Can't really say there is an exact manual that tells you about AutoCAD layout. But, organizations like AIA, Army Corps of Engineers, and a variety of other groups, all have CAD Standards that meet their requirements for drawings. If you are a CAD manager, you can set up AutoCAD to control how your drafters set up drawings and just draw. Meaning if they make a mistake, it tells them what they did wrong. Hope this helps.
The National CAD Standards, including the Uniform Drawing System sections, are probably the closest you'll find to a non-company based document.
You can also refer to NavFac and National Park Service documents. IIRC Harvard Facilities has some on-line as well.
The nice thing about standards is that there are so many of them.
