Does anyone know if it is possible to create a custom material template file that you can load in a new file from a folder?

Sure, Just make a project file with all the materials you want in it, then you can transfer them into your project. Note 2012 lets you know have a custom material library so this isn't really necessary anymore.

Oh ok, thanks that what my plan was, we are just working with multiple template files and want to keep a consistent material library for each file. So far that was the best way I have found to do it. Thanks for you help!

Does this mean I can remove all the Autodesk Material Libraries that are loaded onto users' workstations during deployment? This would help with some of our disk space issues. I never even knew this was a feature of 2012.

No I wouldn't suggest that because you would have to manually repath all the existing materials, unless you weren't going to use any of them.
