I am currently working on the design of an air supported structure with an entry building. Any suggestions on how to model the dome which is 250' W x 420' L x 75' H?

Talk to the manufacturer of your tensile system. At my old firm I worked on a huge tensile structure for a train station, and after a lot of time spent drawing it in a combination of AutoCAD and Microstation, I found out that the tensile structure manufacturer (I think Birdair) had custom in-house software which too care of the whole thing.

If you do draw it in Revit, I would use the massing tool and then assign a curtain grid to it.

Don't go too far without contacting Birdair. From my recollection, if you give them a point cloud, they can connect the dots for you.
