AutoCAD 2016 contains a great many fonts, including "simplex" and "msimplex". I've searched the Googles high and low, and I still don't understand what the differences are between the two. Does anyone have any knowledge to impart unto me?

I'd guess msimplex would be the mono-spaced version of simplex, handy for columns without tabs. Ensures the 15th character on each line will line up with each other.

I thought Msimplex was a monospace font where every character from the narrowest "i" to the widest "m" takes up the same amount of space, while Simplex is a proportional spaced font where each character takes up a different, but appropriate amount of space.

You can see the difference between two fonts by using SHX viewer that you can search "SHX Viewer" in google then you can download that you want.
When I see two fonts, The differences is shape code number but shape word is little same.

Their shxviewer.com website doesn't exist anymore, but there are still places you can download SHX Viewer 1.1 http://freecadapps.mcadcafe.com/swdetails.php?page=category&value=ACG-TDU&orgvalue=ACG&review=4784&rowcolor=fce08d
