Is there a way of creating a variable in an iPart that is user defined? That is the user enters the value when inserting/changing the iPart?

When your in the I-Part Author, have you tried right clicking on the parameter or choice and choosing 'Custom Parameter Column?' This should do what you need, there's also some great tutorials on the subject here (http://www.sdotson.com/tutorials.asp).

So if I understand you correctly, if I have an Extrusion, such as 80-20 or Bosch framing parts, I could model the cross-section of the extrusion with a default length of say 1", then let the user specify an exact length when he inserts it? Would he also be able to "select 2 points of faces" with the measure option and have the extrusion automatically stretch to fit?

I don't think, I-parts would do this, although here's a couple of thoughts... I haven't tried them, but I think they'd work.

1) Place the custom I-part, set the part to adaptive, and use adaptivity to fit the components with assembly constraint.

2) Create an I-feature as a sketch, start the part using that I-feature, then extrude it using the To-From option.

This is possible with a custom iPart.
