I am laying out 2x4 tile ceilings. At least half the time, the direction is opposite of the direction I want. How does one change this to make the 2x4 grid pattern run the other direction?

just tab to one of the grid lines and rotate it

Be aware that if you have ceiling hosted families such as light fixtures, rotating the grid lines will rotate the fixtures as well. This is often fine for small, simple spaces, but can create a problem with larger ceilings hosting many fixtures.

If you are rotating a 2x4 grid wouldn't you want the fixtures to rotate too?

yes, but it will also rotate the fixtures relative to each other, so you end up with fixtures all over the place where they shouldn't be, sometimes even outside the building.
Unless the ceiling direction changes at some point in the design process, you would want to obviously have the ceiling oriented the correct way before inserting fixtures.
