Is there a way to set up a grid line to dimension with centerline tick?

do you mean that you want to see a cl mark instead of a tick? if so, it is not possible. you could create a new dim style that has a different tick style that your typical style.

What I want is for me to dimension the grid lines and the centerline-tick would be automatically applied to it as per the properties settings, not a cl mark.

Unfortunately,
You cannot do this in this. Revit decides what is the "center" of an object and what isn't. So, for instance, it automatically knows that the center of a component should have a centerline tick mark. However, since there is no "centerline" in a grid annotation (it's a line after all) then you can't make this happen.
We like our grid lines to have centerline tick marks as well, and for those PMs who want to use that old CAD standard in Revit, we have a dumb blank component family that we place at the end of the extents of the grid lines close to the head. We place it, align the center to the grid line, and lock the alignment. From then on we dimension to the component instead of to the grid line. Personally, I recommend just dealing with the current limitation under the hopes that Revit will eventually add the option in the properties dialog of a grid line (instance parameter style since some grid lines are for exterior faces of buildings and should get a normal tick mark) to use either the CL or a regular tick.
Then again, I'm the optimistic type...
