Does anyone know how to create a slab that is 12" thick on one side and 11" thick on the opposite side?

Yep, create the slab the thickness you want....then select the slab (I prefer to do it in 3D...you should notice a new toolbar up above. The first button lets you change point elevations. So either add an inch or subtract an inch! :)
Works on flat roofs too....

Make sure you also check "variable" on the concrete layer in the "Edit Assembly" dialog or the slab will just slope from one side to the other with constant thickness.

is this same function you would use to put a depression in your slab?

Probably not... I think it would cause the floor to slope towards the depression instead of just cutting it out. That would be best done with a void family (inplace or not).
