I used a scanned in drawing to pdf and saved it to a TIFF file. I uploaded the TIFF file into a DWG file as an underlay. I need to set up the drawing so that I can get correct scaling for linier measurements.

Pick an image line which is horizontal or vertical and of known length. Assume a horizontal line.
Draw vertical lines at each end of the image line and another line at the proper distance and a horizontal line joining them. With osnap=INTersection use the Relative option of the Scale command to scale the image.

Is this how to set up the scale so that anything drawn over the underlayment will calculate using the LS command? I have drawn many polylines representing computer cabling runs from the server to each user station. I need to find the length of each run using the LS command.

Yes. If you scale to actual size, the polylines should LIST at the correct length in plan view. You may have to consider adding length for elevation changes.
