It's possible, but not the easiest thing to do. It's much easier to work with C3D in VBA or a .NET language.
You might want to look at the C3D Customization group on Autodesk's website. There are a number of posts in there that show how to use Lisp to interact with C3D to do various things.

Thank you for the fast replay.
I am a Civil 3D user. I live in Israel, and we use the metric system.
I need support for various subjects in C3D, but mainly I need solutions in add on applications.
For example I need an application that will select all the sample lines of a certain group name, by selecting either the associated alignment or by selecting one of the sample line.
Please let me know if you can provide this kind of support. or if you know of somebody who can.

The problem is the API. I'll need to check to make sure, but as of C3D 2009, I don't think there's any way to select a Sample Line and identify which sample line it is.
However, there are some tricks that might work. And the API gets better every year, so even if it isn't possible to do exactly what you want in 2009, it might be possible in 2010.
A lot of it depends on exactly what you are doing, why you want to do what you are asking for. Sometimes it's not possible to do one thing, but it's possible to do another thing instead. And of course, sometimes you might discover that if you think about things a little differently, it's possible to do what you want using the built-in functionality of Civil 3D, without any customization at all.
